Twieflingen is situated at an elevation of 103 m south of the nature reserve Elm-Lappwald, about 14 km southwest of the district town Helmstedt. Twieflingen has a population of about 700 (2015) and its part of the municipality Söllingen, which itself if part of the collective municipality Heeseberg (seated in Jerxheim).
The neighbouring villages of Dobbeln and Wobeck were incorporated into Twieflingen in 1974. In 2016, Twieflingen and Ingeleben were incorporated into the municipality of Söllingen.
The church of St. Mauritius dates from the 13th century.
